You can treat both mum and babies for fleas, provided you keep them away from each other until the spray is dry and you keep the spray away from mummy's tummy.

The guidance on Frontline spray has changed recently and you can now treat kittens from a few days old, however you need to get them all down to the vets as Frontline spray is only available with a vet prescription.

I personally think that it is worth the vet visit because everything else you can use in these circumstances works far less well and a bad case of fleas can kill small kittens through anaemia.
